Numeric Format:  (d* function)/(d varaible = point), press Enter
In RPN Mode: You also need to press Shift+Comma (Eval)
Symbolic Format (CAS Mode):  (d function)/(d variable)

*d represents the derivative symbol

diff command:
diff(function, variable)
